== Etymology == The word analgesic derives from Greek an- (ἀν-, "without"), álgos (ἄλγος, "pain"), and -ikos (-ικος, forming adjectives). Such drugs were usually known as "anodynes" before the 20th century.
Foreign versions of Lipitor and Celebrex were smuggled into the U.S. from South America and resold after being repackaged to conceal the true origin of the drugs. Counterfeit Lipitor was manufactured in South America and smuggled into the US, where it sold after commingling with the genuine foreign Lipitor. Participants conspired to buy, sell and traffic almost eight million dollars' worth of stolen Glaxo Smith Kline and Roche drugs, using fake pedigrees to launder the drugs and thereby concealing that they were stolen. There also were charges related to the sale of counterfeit Procrit, as well as counterfeit and misbranded Serostim and Neupogen. Procrit is an injectable drug used in the treatment of anemia, Serostim is Serono's brand name form of synthetic somatropin (i.e. human growth hormone formed using laboratory methods of genetic recombination) marketed for HIV-associated wasting, and Neupogen is an injectable drug used by cancer patients to stimulate the production of white blood cells in order to decrease the incidence of infections. In 2005, the FDA held a Congressional hearing to review the situation. The U.S. is an especially attractive market for counterfeiters, because 40% of worldwide annual prescription drug sales were made in the United States in 2007. In 2011, a "PROTECT IP Act" was proposed to deter advertising. == Signs and symptoms == The typical screening process for sleep apnea involves asking patients about common symptoms such as snoring, witnessed pauses in breathing during sleep, and excessive daytime sleepiness. There is a wide range in presenting symptoms in patients with sleep apnea, from being asymptomatic to falling asleep while driving. Due to this wide range in clinical presentation, some people are not aware that they have sleep apnea and are either misdiagnosed or ignore the symptoms altogether. A current area requiring further study involves identifying different subtypes of sleep apnea based on patients who tend to present with different clusters or groupings of particular symptoms. OSA may increase the risk of car crashes and work-related injuries due to sleep fragmentation from repeated arousals during sleep. If OSA is not treated, it results in excessive daytime sleepiness and oxidative stress from the repeated drops in oxygen saturation, and people are at increased risk of other systemic health problems, such as diabetes, hypertension or cardiovascular disease. Subtle manifestations of sleep apnea may include treatment refractory hypertension and cardiac arrhythmias and over time as the disease progresses, more obvious symptoms may become apparent. Due to the disruption in daytime cognitive state, behavioral effects may be present. These can include moodiness, belligerence, as well as a decrease in attentiveness and energy. === Severe MTHFR deficiency === Severe MTHFR deficiency is rare (about 50 cases worldwide) and caused by mutations resulting in 0–20% residual enzyme activity. Patients exhibit developmental delay, motor and gait dysfunction, seizures, and neurological impairment and have extremely high levels of homocysteine in their plasma and urine as well as low to normal plasma methionine levels. This deficiency and mutations in MTHFR have also been linked to recessive spastic paraparesis with complex I deficiency. A study on the Chinese Uyghur population indicated that rs1801131 polymorphism in MTHFR was associated with nsCL/P in Chinese Uyghur population. Given the unique genetic and environmental characters of the Uyghur population, these findings may be helpful for exploring the pathogenesis of this complex disease.
=== 1949–1950 === Crick taught himself the mathematical theory of X-ray crystallography. During the period of Crick's study of X-ray diffraction, researchers in the Cambridge lab were attempting to determine the most stable helical conformation of amino acid chains in proteins (the alpha helix). Linus Pauling was the first to identify the 3.6 amino acids per helix turn ratio of the alpha helix. Crick was witness to the kinds of errors that his co-workers made in their failed attempts to make a correct molecular model of the alpha helix; these turned out to be important lessons that could be applied, in the future, to the helical structure of DNA. For example, he learned the importance of the structural rigidity that double bonds confer on molecular structures which is relevant both to peptide bonds in proteins and the structure of nucleotides in DNA.
== Medical uses == Ixazomib is used in combination with lenalidomide and dexamethasone for the treatment of multiple myeloma in adults after at least one prior therapy. There are no experiences with children and youths under 18 years of age. The study relevant for approval included 722 people. In this study, ixazomib increased the median time of progression-free survival from 14.7 months (in the placebo+lenalidomide+dexamethasone study arm including 362 people) to 20.6 months (under ixazomib+lenalidomide+dexamethasone, 360 people), which was a statistically significant effect (p = 0.012). 11.7% of patients in the ixazomib group had a complete response to the treatment, versus 6.6% in the placebo group. Overall response rate (complete plus partial) was 78.3% versus 71.5%. A phase 3 study demonstrated a significant improvement in progression-free survival (PFS) with ixazomib-lenalidomide-dexamethasone (IRd) compared with placebo. High-risk cytogenetic abnormalities were defined as del(17p), t(4;14), and/or t(14;16); additionally, patients were assessed for 1q21 amplification. Of 722 randomized patients, 552 had cytogenetic results; 137 (25%) had high-risk cytogenetic abnormalities and 172 (32%) had 1q21 amplification alone. PFS was improved with IRd versus placebo in both high-risk and standard-risk cytogenetics subgroups: in high-risk patients, with median PFS of 21.4 versus 9.7 months; in standard-risk patients, with median PFS of 20.6 versus 15.6 months.

Weighted Companion Cubes are variants of Weighted Storage Cubes that are used in the testing chambers. They are differentiated from regular Storage Cubes with small pink hearts on its external surface, instead of an Aperture Science logo. In Portal, during test chamber 17, Chell is given a Weighted Companion Cube by GLaDOS, told by her that it will be her faithful companion for the duration of the test. Chell must use the Cube to complete the chamber, at which point GLaDOS requires her to incinerate the Cube before being able to progress further. The requirement to incinerate the companion cube originates from test-play of Portal, during which players brought their companion cube with them to later levels, with unexpected impact on game play. Another Companion Cube is seen at the end of the game, standing next to the cake. In Portal 2, redesigned Weighted Companion Cubes appear in test chamber 7 in chapter 2. GLaDOS fizzles two of them at the start of the test to annoy Chell. Chell is given the option to take a Weighted Companion Cube with her at the end of the test, but if Chell does this, GLaDOS fizzles the Companion Cube before Chell can enter the lift leading to the next test chamber. A Companion Cube with the old design, supposedly the one from Portal, appears at the very end of the game, as it is ejected, charred and singed, from the shed (possibly at ground level) shortly after Chell's departure. Companion Cubes are also mentioned a lot in the Ratman dens in Portal. In the Lab Rat comic, Ratman is also accompanied by a Companion Cube, which he imagines talks to him.
== Renal Impairment == Dose adjustment is necessary based on the degree of renal function: Mild Renal Impairment (eGFR ≥ 60 mL/min/1.73 m²): No dose adjustment required. Moderate Renal Impairment (eGFR 30–59 mL/min/1.73 m²): Reduce dose to 50 mg once weekly. Severe Renal Impairment or ESRD (eGFR < 30 mL/min/1.73 m² or on dialysis): Reduce dose to 25 mg once weekly, with close monitoring.
In 1987, according to Department of State and congressional staff, drug traffickers were offering UMOPAR officers and town officials in the Chapare region amounts ranging from US$15,000 to US$25,000 for seventy two hours of "protection" in order to allow aircraft to load and take off from clandestine airstrips. In February 1988, the deputy minister of national defense announced that about 90 percent of UMOPAR members, including twelve middle- and high-ranking officers, had been dismissed for alleged links to drug trafficking. The La Paz newspaper Presencia reported in March 1988 that UMOPAR chiefs, including the prosecutors, were working with narcotics traffickers by returning to them the large drug finds and turning only the small ones in to the authorities. Observers considered UMOPAR forces in Santa Cruz to be more honest and dedicated. In October 1988, the undersecretary of the Social Defense Secretariat reiterated that drug traffickers had obtained the protection of important sectors of influence in Bolivia, including some military members and ordinary judges. He cited the example of Cochabamba's Seventh Division commander and four of his top officers, who were discharged dishonorably after they were found to be protecting a clandestine Chapare airstrip used by drug smugglers. The ministry official also announced that the navy was protecting drug-trafficking activities in the Puerto Villarroel area of the Chapare. For that reason, the United States suspended assistance to the navy temporarily in late 1988 until its commander was replaced.
