== Columns == The columns used in FPLC are large (inner diameters on the order of millimeters) tubes that contain small (micrometer-scale) particles or gel beads as the stationary phase. The chromatographic bed is composed of gel beads inside the column and the sample is introduced into the injector and carried into the column by the flowing solvent. As a result of different components adhering to or diffusing through the gel, the sample mixture gets separated. Columns used with an FPLC can separate macromolecules based on size (size-exclusion chromatography), charge distribution (ion exchange), hydrophobicity, reverse-phase or biorecognition (as with affinity chromatography). For easy use, a wide range of pre-packed columns for techniques such as ion exchange, gel filtration (size exclusion), hydrophobic interaction, and affinity chromatography are available. FPLC differs from HPLC in that the columns used for FPLC can only be used up to maximum pressure of 3-4 MPa (435-580 psi). Thus, if the pressure of HPLC can be limited, each FPLC column may also be used in an HPLC machine.

=== Dez Watkins === Dez Watkins (Martin Hancock) is a fixer working on behalf of Russian foreign intelligence to protect the interests of Tender. He is first seen approaching Rishi Ramdani and financial journalist Jim Dycker at a bar, warding off an apparent scammer. Rishi and Dycker invite Watkins for drinks and go back to Rishi's apartment to do drugs, where Dycker declares his plan to continue investigating Tender despite being fired by his publication. Watkins supplies them cocaine laced with fentanyl and raises the volume on Rishi's stereo before leaving, inciting a noise complaint that draws police to the apartment right as Dycker overdoses and dies from the poisoned drugs; Rishi is arrested while attempting to flee and is subsequently charged with manslaughter. Watkins later stops Whitney from attempting to disappear amidst escalating public scrutiny of Tender's fraud, having tracked him down to New York after Whitney's burner phone connected to airport WiFi. Watkins and Tender board member Ferdinand Schwarzwald (also a Russian asset) force Whitney to proceed with a takeover bid for Pierpoint, and warn Whitney that escape is not an option, with Watkins confirming that he murdered Dycker.

== Structure == LRP5 is a transmembrane low-density lipoprotein receptor that shares a similar structure with LRP6. In each protein, about 85% of its 1600-amino-acid length is extracellular. Each has four β-propeller motifs at the amino terminal end that alternate with four epidermal growth factor (EGF)-like repeats. Most extracellular ligands bind to LRP5 and LRP6 at the β-propellers. Each protein has a single-pass, 22-amino-acid segment that crosses the cell membrane and a 207-amino-acid segment that is internal to the cell.
The constitutional status of a union territory is similar to a state under the perennial president's rule per Article 356 subject to specific exemptions to a few union territories with legislative assembly. As Per Article 240 (1), supreme power is accorded to the president in regulating the affairs of all the union territories except Chandigarh, NCT and Puducherry, including powers to override the laws made by Parliament and the constitution of India. Article 240(2) allows implementing tax haven laws in these union territories to attract foreign capital and investments into India instead of depending on foreign tax haven countries. == Signaling pathway == The essential function of the TCR complex is to identify specific bound antigen derived from a potentially harmful pathogen and elicit a distinct and critical response. At the same time it has to ignore any self-antigen and tolerate harmless antigens such as food antigens. The signal transduction mechanism by which a T cell elicits this response upon contact with its unique antigen is termed T-cell activation. Upon binding to pMHC, the TCR initiates a signaling cascade, involving transcription factor activation and cytoskeletal remodeling resulting in T-cell activation. Active T cells secrete cytokines, undergo rapid proliferation, have cytotoxic activity and differentiate into effector and memory cells. When the TCR is triggered, T cells form an immunological synapse allowing them to stay in contact with the antigen presenting cell for several hours. On a population level, T-cell activation depends on the strength of TCR stimulation, the dose–response curve of ligand to cytokine production is sigmoidal. However, T-cell activation on a single cell level can be characterized by a digital switch-like response, meaning the T cell is fully activated if the stimulus is higher than a given threshold; otherwise the T cell stays in its non-activated state.
