Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Two structural changes distinguish the synthetic peptide from the natural hormone. A norleucine residue replaces methionine at one position, and a D-configured phenylalanine replaces the natural L-form at another. Both substitutions slow enzymatic breakdown, which extends the molecule's persistence relative to the parent hormone. The lactam bridge further constrains the backbone into a stable conformation. These features are standard design strategies in peptide chemistry and are not unique to this compound; they appear across many research peptides built for improved stability.
The compound was developed in the late 1980s and early 1990s by academic researchers investigating melanocortin signaling and pigmentation. Early work explored whether synthetic analogs could reproduce effects of the natural hormone under controlled conditions. The molecule never advanced through the full regulatory pathway required for approval as a medicine. From the mid-2000s onward it appeared in unregulated consumer markets, often distributed through informal channels. That gap between research origins and commercial availability shapes how the compound is discussed today.
Reversed-phase high-performance liquid chromatography is the routine method for purity assessment. Peptides absorb near 214 nm because of the peptide bond, and a gradient of acetonitrile in water separates the intact peptide from deletion sequences, oxidised products, and earlier-eluting fragments at neutral pH. Electrospray ionisation mass spectrometry provides an orthogonal check: the measured mass must agree with the theoretical value. Amino acid analysis and peptide mapping confirm structure but are used less often. Reference standards remain scarce because the peptide is not described in any pharmacopoeia.
Regulatory treatment varies by country. In the United States the peptide is not approved as a medicine, and products offered for human use may be treated as unapproved new drugs; some states also restrict sale. Australia, the United Kingdom, and European Union member states apply comparable restrictions to unapproved peptide products. Border agencies have seized shipments labelled as research chemicals. Classification may change over time, and the legal position for personal importation is not clearly settled in most published guidance.
Lyophilised melanotan-2 is supplied as a solid, which is more stable than a solution. The material is hygroscopic, so weighing is done quickly, in low humidity, with the container kept sealed. Reconstitution usually uses water for injection or bacteriostatic water, added down the wall of the vial to limit foaming. A reconstituted solution is held at 2 to 8 °C and kept away from light. Repeated freezing and thawing of the same vial is avoided because ice crystal formation and concentration effects degrade the peptide.

Outside regulated medicine, melanotan II circulates through online vendors as a research chemical, often marketed for tanning. Products sold this way vary widely in purity, concentration, and labeling accuracy, and independent testing has documented discrepancies. Published reports describe both pigment effects and adverse reactions, including nausea, flushing, and darkening of existing moles. Long-term safety data are sparse, and no large controlled trial has established a risk profile. Questions about cumulative effects on melanocytes remain unresolved in the literature.
Melanotan II is a synthetic peptide analog modeled on alpha-melanocyte-stimulating hormone, a naturally occurring signaling peptide involved in pigmentation. Its structure is a cyclic heptapeptide containing two non-natural substitutions, norleucine at position four and D-phenylalanine at position seven. These modifications resist enzymatic breakdown and extend the molecule's activity relative to the native hormone. The compound binds melanocortin receptors and is studied mainly as a pharmacological tool rather than a therapeutic product. It has never received approval as a medicine in any major jurisdiction.
The compound was developed in the late 1980s and 1990s by academic researchers investigating photoprotection. The rationale held that stimulating melanin production might reduce ultraviolet damage to skin and lower skin cancer risk. Early work examined receptor binding, pigment response, and short-term tolerability in small studies. That program did not produce an approved drug, and formal development stalled after early-phase trials. Whether induced pigmentation confers meaningful photoprotection remains an open question.
Verification of a purchased sample requires documentation linking a batch to a certificate of analysis, and that document should be read for the methods used rather than the headline purity figure. A single chromatographic percentage does not establish identity. Independent laboratories can perform identity and content assays, but no such test establishes that a product is suitable for human use. Claims about efficacy rest largely on small, early studies rather than on replicated controlled trials, and that gap remains open.
Identity testing for a cyclic peptide of this size usually relies on reversed-phase high-performance liquid chromatography coupled to mass spectrometry. The mass spectrum confirms molecular weight, while the chromatographic trace indicates the proportion of related impurities. Tandem mass spectrometry can provide sequence-level information when fragmentation data are compared against a reference standard. Nuclear magnetic resonance is sometimes used to confirm the lactam bridge, although it requires more material and greater operator expertise than routine chromatographic methods.
It is also expected that thorium-232 should be able to undergo double beta decay, which would produce uranium-232, but this has not yet been observed experimentally. All isotopes from 232U to 236U inclusive have minor cluster decay branches (less than 10−10%), and all these bar 233U, in addition to 238U, have minor spontaneous fission branches; the greatest branching ratio for spontaneous fission is about 5×10−5% for 238U, or about one in every two million decays. The shorter-lived trace isotopes 237U and 239U exclusively undergo beta decay, with respective half-lives of 6.752 days and 23.45 minutes. In total, 28 isotopes of uranium have been identified, ranging in mass number from 214 to 242, with the exception of 220. Among the uranium isotopes not found in natural samples or nuclear fuel, the longest-lived is 230U, an alpha emitter with a half-life of 20.23 days. This isotope has been considered for use in targeted alpha-particle therapy (TAT). All other isotopes have half-lives shorter than one hour, except for 231U (half-life 4.2 days) and 240U (half-life 14.1 hours). The shortest-lived known isotope is 221U, with a half-life of 660 nanoseconds, and it is expected that the hitherto unknown 220U has an even shorter half-life. The proton-rich isotopes lighter than 232U primarily undergo alpha decay, except for 229U and 231U, which decay to protactinium isotopes via positron emission and electron capture, respectively; the neutron-rich 240U, 241U, and 242U undergo beta decay to form neptunium isotopes.
=== Parallel approach === Parallel engineering is also known as bioengineering. The basic genetic code is the foundation for parallel engineering research, which uses conventional biomolecules like nucleic acids and the 20 amino acids to construct biological systems. For a variety of applications in biocomputing, bioenergy, biofuels, bioremediation, optogenetics, and medicine, it involves the standardisation of DNA components, engineering of switches, biosensors, genetic circuits, logic gates, and cellular communication operators. For directing the expression of two or more genes and/or proteins, the majority of these applications often rely on the use of one or more vectors (or plasmids). Small, circular, double-strand DNA units known as plasmids, which are primarily found in prokaryotic but can also occasionally be detected in eukaryotic cells, may replicate autonomously of chromosomal DNA.
P. porrigens was once generally regarded as edible, though bland. As of 2011, it is a suspect in two outbreaks in Japan involving fatal encephalopathy. Most victims had preexisting kidney disorders. The first incident occurred in September and October 2004 across nine prefectures in Japan, documenting the sickening of 59 people and the eventual death of 17. Most of those who died had preexisting liver problems and the average age of those affected was 70. Death occurred between 13 and 29 days after the onset of symptoms, which occurred at most three weeks after consumption of the species. The second incident occurred in 2009, when a 65-year-old man who had been on hemodialysis died from acute encephalopathy after eating P. porrigens. The mechanism of action for the toxicity of P. porrigens has not been definitively established, but several possibilities have been suggested. It has been demonstrated that P. porrigens contains an unusual amino acid, Pleurocybellaziridin, which is toxic to the brain cells of rats in cell culture studies, but it has not yet been possible to definitively determine that this was the cause of the fatal encephalopathies. Other mechanisms have been suggested for P. porrigens's apparent toxicity, including the possibility that the fungus may contain toxic levels of cyanide salts. A proposed mechanism of action for the toxicity of P. porrigens has been proposed by Kawagishi, et al.
A polyhistidine-tag, best known by the trademarked name His-tag, is an amino acid motif in proteins that typically consists of at least six histidine (His) residues, often at the N- or C-terminus of the protein. It is also known as a hexa histidine-tag, 6xHis-tag, or His6 tag. The tag was invented by Roche, although the use of histidines and its vectors are distributed by Qiagen. Various purification kits for histidine-tagged proteins are commercially available from multiple companies. The total number of histidine residues may vary in the tag from as low as two, to as high as 10 or more His residues. N- or C-terminal His-tags may also be followed or preceded, respectively, by a suitable amino acid sequence that facilitates removal of the polyhistidine-tag using endopeptidases. This extra sequence is not necessary if exopeptidases are used to remove N-terminal His-tags (e.g., Qiagen TAGZyme). Furthermore, exopeptidase cleavage may solve the unspecific cleavage observed when using endoprotease-based tag removal. Polyhistidine-tags are often used for affinity purification of genetically modified proteins.

==== Memories of childhood ==== Jung was a solitary and introverted child. From childhood, he believed that, like his mother, he had two personalities—a modern Swiss citizen and a personality more suited to the 18th century. "Personality Number 1", as he termed it, was a typical schoolboy living in the era of the time, his conscious personality. "Personality Number 2" was a dignified, authoritative, and influential man from the past, his unconscious. Though Jung was close to both parents, he was disappointed by his father's academic approach to faith.
Saeujeot (새우젓) or myeolchijeot is not added to the kimchi spice-seasoning mixture, but is simmered first to reduce odors, eliminate tannic flavor and fats, and then is mixed with a thickener made of rice or wheat starch (풀). This technique has been falling into disuse in the past 40 years.
Further developments in Poland occurred concurrently with and were influenced by the reformist leadership of Mikhail Gorbachev in the Soviet Union (processes known as Glasnost and Perestroika). In September 1986, a general amnesty was declared and the government released nearly all political prisoners. However, the country lacked basic stability, as the regime's efforts to organize society from the top down had failed, while the opposition's attempts at creating an "alternate society" were also unsuccessful. With the economic crisis unresolved and societal institutions dysfunctional, both the ruling establishment and the opposition began looking for ways out of the stalemate. Facilitated by the indispensable mediation of the Catholic Church, exploratory contacts were established. Student protests resumed in February 1988. Continuing economic decline led to strikes across the country in April, May and August. The Soviet Union, increasingly destabilized, was unwilling to apply military or other pressure to prop up allied regimes in trouble. The Polish government felt compelled to negotiate with the opposition and in September 1988 preliminary talks with Solidarity leaders ensued in Magdalenka. Numerous meetings that took place involved Wałęsa and General Kiszczak, among others. In November, the regime made a major public relations mistake by allowing a televised debate between Wałęsa and Alfred Miodowicz, chief of the All-Poland Alliance of Trade Unions, the official trade union organization.
Chain-ganging occurs when a state sees its own security tied to the security of its alliance partner. It chains itself by deeming any attack on its ally the equivalent of an attack on itself. That is another aspect of the balance of power theory, whereby the smaller states could drag their chained states into wars that they have no desire to fight. A key example was the chain-ganging between states prior to World War I, dragging most of Europe to war over a dispute between the relatively major power of Austria-Hungary and the minor power of Serbia. Thus, states "may chain themselves unconditionally to reckless allies whose survival is seen to be indispensable to the maintenance of the balance".

=== Biosynthesis === It is believed that the biosynthesis of guanacastepenes occurs via the mevalonate pathway. This pathway begins with acetyl-CoA and yields both isopentenyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP). IPP and DMAPP are then converted into geranylgeranyl pyrophosphate (GGPP), the most important diterpene precursor. A cationic cyclization cascade converts GGPP into the macrocyclic intermediate β-araneosene and ultimately into the dolabellane skeleton. Dolabellane is then rearranged into the neodolabellane skeleton via stereospecific hydride and methyl shifts, thereby fixing the stereochemistry at C11 and C12 and shifting the C15 methyl group across the ring fusion. The carbon skeleton of Guanacastepene A is closely related to the dolastane and neodolabellane families. Therefore, it is assumed that the biogenesis of dolastanes proceeds via a further intramolecular cyclization of the dolabellane-derived cation, resulting in the tricyclic [5-7-6]-dolastane (guanacastane) skeleton. This basic framework is diversified through a series of oxidation reactions in which the characteristic functional groups of guanacastepenes are incorporated. The guanacastepenes characterized to date represent only a fraction of the metabolites present in the CR115 extract. The tricyclic guanacastepenes A, B, and C are considered the simplest members of this family; successive oxidation and functionalization are thought to lead to the formation of structurally more complex ring systems.
== Limitations == Because the Edman degradation proceeds from the N-terminus of the protein, it will not work if the N-terminus has been chemically modified (e.g. by acetylation or formation of pyroglutamic acid). Sequencing will stop if a non-α-amino acid is encountered (e.g. isoaspartic acid), since the favored five-membered ring intermediate is unable to be formed. Edman degradation is generally not useful to determine the positions of disulfide bridges. Protein sequencing of attomole level of Edman degraded sequences are obtainable but require accelerator mass spectrometery which requires large, complex, and expensive equipment as well as 30 hours of bench time to analyze a single run.
The oil industry remained largely state-owned, with the wholly state-owned National Oil Corporation retaining a 70% share in Libya's oil industry, the government also imposed a 93% tax on all oil that foreign companies produced in Libya. Price controls and subsidies over oil and food remained in place, and state-provided benefits such as free education, universal healthcare, free housing, free water and free electricity remained in place. Libya also changed its stance on the WTO after the removal of technocrat Shukri Ghanem, with Gaddafi condemning the WTO as a neocolonial terrorist organisation, and urging African and Third World countries not to join it. While there was no accompanying political liberalization, with Gaddafi retaining predominant control, in March 2010, the government devolved further powers to the municipal councils. Rising numbers of reformist technocrats attained positions in the country's governance; best known was Gaddafi's son and heir apparent Saif al-Islam Gaddafi, who was openly critical of Libya's human rights record. He led a group who proposed the drafting of a new constitution, although it was never adopted. Involved in encouraging tourism, Saif founded several privately run media channels in 2008, but after criticizing the government, they were nationalized in 2009.
Intrinsic disorder can be either annotated from experimental information or predicted with specialized software. Disorder prediction algorithms can predict intrinsic disorder (ID) propensity with high accuracy (approaching around 80%) based on primary sequence composition, similarity to unassigned segments in protein x-ray datasets, flexible regions in NMR studies and physico-chemical properties of amino acids.
The 1980s MGN W12 F1 engine used rotary valves but never raced. Between 2002 and 2004 the Australian developer Bishop Innovation and Mercedes-Ilmor tested rotary valves for a F1 V10 engine. Bishop Innovations' patent for the rotary valve engine was bought out by BRV Pty Ltd, owned by Tony Wallis, one of the valves original designers. BRV has constructed several functional motors using the rotary valve technology, such as a Honda CRF 450, which had greater torque at both low (17% increase) and high (9% increase) engine speeds, and also produced more brake horsepower up to around 30% more at functional engine speeds. The engine was also considerably smaller and lighter, as the cylinder head assembly was not as large. A company in the UK called Roton Engine Developments made some progress in 2005 with a two-rotor (one for inlet and one for exhaust) single-cylinder Husaberg motorcycle engine. They filed patents and got an example running in 2006, but were backed by MG Rover which subsequently went bust, leaving Roton without enough funds to continue. The designs surfaced some years later in Australia with Engine Developments Australia Pty Ltd. A prototype casting was produced in 2013 on a Kawasaki Ninja 300 parallel twin unit.

It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
Melanotan-1, also called afamelanotide, is a shorter linear analog with a different amino acid sequence and no lactam ring. It has been evaluated in formal clinical programs, while melanotan-2 has not. The two are distinct molecules and are not interchangeable.
No. Bremelanotide is a related but distinct cyclic peptide that reached approved status for one specific clinical indication. Melanotan-2 is a separate molecule with its own sequence and properties. Shared ancestry in the melanocortin family does not make them the same substance.
Low temperature slows hydrolysis and oxidation, the two main routes by which the peptide backbone and side chains are modified. A lyophilised powder stored at −20 °C is more stable than one kept at room temperature, and once the material is dissolved the degradation rate rises, making refrigeration more important.
