Melanotan-2 is a synthetic peptide designed as an analog of alpha-melanocyte-stimulating hormone, a signaling molecule produced in the pituitary and skin. Its structure is a linear chain of seven amino acids that folds into a ring through an internal lactam bridge joining two side chains. The compound is sometimes written as MT-II or MEL-2 in informal and commercial contexts. It belongs to the melanocortin peptide family, a group of short signaling molecules that share a conserved core sequence recognized by melanocortin receptors.
Two structural changes distinguish the synthetic peptide from the natural hormone. A norleucine residue replaces methionine at one position, and a D-configured phenylalanine replaces the natural L-form at another. Both substitutions slow enzymatic breakdown, which extends the molecule's persistence relative to the parent hormone. The lactam bridge further constrains the backbone into a stable conformation. These features are standard design strategies in peptide chemistry and are not unique to this compound; they appear across many research peptides built for improved stability.
Receptor studies place melanotan-2 among non-selective melanocortin agonists, binding MC1R, MC3R, MC4R and MC5R rather than a single subtype. Activation of MC1R on cutaneous melanocytes raises tyrosinase activity and shifts pigment synthesis toward eumelanin, which is darker and more photostable than pheomelanin. Central receptors, particularly MC4R, are associated with appetite suppression and with reported effects on sexual function. Because subtype selectivity is low, the same molecule engages pigment, metabolic and vascular pathways at once, and this breadth is a common explanation offered for the range of adverse events described in user reports.
No regulatory authority has approved melanotan-2 for human use, and several countries classify it as a prescription-only or controlled substance, which restricts lawful supply. Material sold online is generally labelled as a research chemical and is not required to meet pharmaceutical standards of identity or purity. Published human data consist mainly of small uncontrolled studies, case reports and adverse-event notifications, so the evidence base is descriptive rather than confirmatory. Whether repeated melanocyte stimulation alters long-term naevus behaviour remains an open question that no completed trial has resolved.
| Property | Value | Notes |
|---|---|---|
| Molecular class | Synthetic cyclic heptapeptide | Analog of alpha-MSH with an internal lactam bridge |
| Molecular weight | Approximately 1024 daltons | Free base value; salt forms differ |
| Appearance | White to off-white powder | Usually supplied as a lyophilized solid |
| Solubility | Freely soluble in water | Poorly soluble in nonpolar solvents |
| Typical storage | −20 °C, dry, protected from light | Repeated freeze-thaw cycles degrade peptides |
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.
Early published reports described melanotan-2 as a tanning agent without sun protection, which means darkening is not the same as protection against ultraviolet radiation. Later studies explored the peptide in erectile dysfunction, hemorrhagic shock, and some skin conditions. No regulator in the United States or Europe has approved it for clinical use. Many products labelled melanotan-2 are sold without approval and their identity and purity are unverified. Its long-term safety in humans remains an open question.
Melanotan-2, also written Melanotan II, is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone. Its sequence is Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, and the lactam bridge between the aspartate and lysine side chains constrains the peptide into a ring. This structural change increases receptor affinity and metabolic stability relative to the native hormone. The compound was created in the 1980s as a research tool for studying pigmentation biology.
== Enzyme activity == Activated by calcium, the enzyme digests proteins preferentially after hydrophobic amino acids (aliphatic, aromatic and other hydrophobic amino acids). Although calcium ions do not affect the enzyme activity, they do contribute to its stability. Proteins will be completely digested if the incubation time is long and the protease concentration high enough. Upon removal of the calcium ions, the stability of the enzyme is reduced, but the proteolytic activity remains. Proteinase K has two binding sites for Ca2+, which are located close to the active center, but are not directly involved in the catalytic mechanism. The residual activity is sufficient to digest proteins, which usually contaminate nucleic acid preparations. Therefore, the digestion with Proteinase K for the purification of nucleic acids is usually performed in the presence of EDTA (inhibition of metal-ion dependent enzymes such as nucleases). Proteinase K is also stable over a wide pH range (4–12), with a pH optimum of pH 8.0. An elevation of the reaction temperature from 37 °C to 50–60 °C may increase the activity several times, like the addition of 0.5–1% sodium dodecyl sulfate (SDS) or Guanidinium chloride (3 M), Guanidinium thiocyanate (1 M) and urea (4 M) . The above-mentioned conditions enhance proteinase K activity by making its substrate cleavage sites more accessible. Temperatures above 65 °C, trichloroacetic acid (TCA) or the serine protease-inhibitors AEBSF, PMSF or DFP inhibit the activity.
== Etymology == The Oxford English Dictionary had cited the first known usage in the English language to a Scottish newspaper, The People's Journal, in 1848: "A war among the great powers is now necessarily a world-war." The term "world war" is used by Karl Marx and his associate, Friedrich Engels, in a series of articles published around 1850 called The Class Struggles in France. Rasmus B. Anderson in 1889 described an episode in Teutonic mythology as a "world war" (Swedish: världskrig), justifying this description by a line in an Old Norse epic poem, "Völuspá: folcvig fyrst I heimi" ("The first great war in the world"). German writer August Wilhelm Otto Niemann used the term "world war" in the title of his anti-British novel, Der Weltkrieg: Deutsche Träume (The World War: German Dreams) in 1904, published in English as The Coming Conquest of England. The term "first world war" was first used in September 1914 by German biologist and philosopher Ernst Haeckel, who claimed that "there is no doubt that the course and character of the feared 'European War' ... will become the first world war in the full sense of the word", citing a wire service report in the Indianapolis Star on 20 September 1914. In English, the term "First World War" was used by Lieutenant Colonel Charles à Court Repington as the title of his memoirs, published in 1920; he had previously noted his discussion on the matter with a Major Johnstone of Harvard University in his diary entry of September 10, 1918. The term "World War I" was coined by Time magazine on page 28 of its June 12, 1939, issue.
The Ocean Race is a yacht race around the world, held every three or four years since 1973. Originally named the Whitbread Round the World Race after its initiating sponsor, British brewing company Whitbread, in 2001 it became the Volvo Ocean Race after Swedish automobile manufacturer Volvo took up the sponsorship, and in 2019 it was renamed The Ocean Race. Each of the entries has a sailing crew who race day and night for more than 20 days at a time on some of the legs. Since the 2008–2009 race there has also been a dedicated media crew member called the On Board Reporter (OBR), who does not contribute to the sailing of the boat, but is responsible for sending images and video to race headquarters via satellite from the middle of the ocean. In the 2017–2018 race, the number of crew ranged between 7 and 10 (plus the OBR) depending on the gender ratio, with the rules providing an incentive to having women sailors on board. To minimize weight, crews overwhelmingly rely on freeze-dried fare for sustenance. Crews are also subjected to temperature variations from −5 to +40 degrees Celsius and will often take only one change of clothes.
HS(CH2)2SH + 2 NR3 + 2 R'I → R'S(CH2)2SR' + 2 [R3NH]I Oxidation of 1,2-ethanedithiol gives a series of oligomers, including the cyclic bis(disulfide). As a 1,2-dithiol, this compound reacts with aldehydes and ketones to give 1,3-dithiolanes, which can be useful intermediates.

== Inside the body == Hemolysis inside the body can be caused by a large number of medical conditions, including some parasites (e.g., Plasmodium), some autoimmune disorders (e.g., autoimmune haemolytic anaemia), drug-induced hemolytic anemia, atypical hemolytic uremic syndrome (aHUS)), some genetic disorders (e.g., Sickle-cell disease or G6PD deficiency), or blood with too low a solute concentration (hypotonic to cells). Hemolysis can lead to hemoglobinemia due to hemoglobin released into the blood plasma, which plays a significant role in the pathogenesis of sepsis and can lead to increased risk of infection due to its inhibitory effects on the innate immune system.
== Gameplay == As with previous Half-Life games, Episode Two is played in the first person as Gordon Freeman against transhuman troops, known as the Combine, and other hostile alien creatures. Levels are linear but add a more open environment, consisting of puzzles and first-person shooter (FPS) gameplay. Sequences involving vehicles are interspersed throughout the game, breaking up moments of combat. One of the focal points was the increased use of vehicles in open areas. However, the game retains its original linear style until the final battle. Episode Two has more puzzles than Episode One; the sequence in which the player must cross a damaged bridge was the largest physics puzzle in the series up to that point. As in the previous two games, Episode Two features numerous "achievements" (similar to PlayStation 3's Trophies and Xbox Live's Achievements) for carrying out certain tasks. Some are essential to game progress, such as helping fight off an antlion invasion, or defeating the first Hunters. Others are optional tricks or feats the player can perform, such as killing a Combine soldier with their own grenade or running down a certain number of enemies with the car. Episode Two features a new enemy, the Hunter, which is seen briefly in a recorded message in Episode One. The Hunter is one of the most dangerous enemies and serves as means of emotional development for Alyx Vance. The Hunter is a powerful and resilient enemy which players must often run from while seeking a means to fight back; Episode Two's environments are designed with this in mind.
Because the formation of PTH regulates the calcium level in the blood, it can affect all areas of the body. The overactivity of a parathyroid gland is known as hyperparathyroidism. It is unknown what directly causes hyperparathyroidism. However there are many factors that can cause over-secretion of PTH. The further consequence of this disorder can be osteopenia, or even osteoporosis, which is the loss of bone density. This leaves bones more porous, fragile, and likely to experience fracture. This can be detected by usage of dual-energy X-ray absorptiometry (DEXA). Interesting enough, a derivative of synthetic PTH is often given to patients with osteoporosis to combat the disease.

The basic components of a mature mammary gland are the alveoli (hollow cavities, a few millimeters large), which are lined with milk-secreting cuboidal cells and surrounded by myoepithelial cells. These alveoli join to form groups known as lobules. Each lobule has a lactiferous duct that drains into openings in the nipple. The myoepithelial cells contract under the stimulation of oxytocin, excreting the milk secreted by alveolar units into the lobule lumen toward the nipple. As the infant begins to suck, the oxytocin-mediated "let down reflex" ensues, and the mother's milk is secreted—not sucked—from the gland into the infant's mouth. All the milk-secreting tissue leading to a single lactiferous duct is collectively called a "simple mammary gland"; in a "complex mammary gland", all the simple mammary glands serve one nipple. Humans normally have two complex mammary glands, one in each breast, and each complex mammary gland consists of 10–20 simple glands. The opening of each simple gland on the surface of the nipple is called a "pore." The presence of more than two nipples is known as polythelia and the presence of more than two complex mammary glands as polymastia. Maintaining the correct polarized morphology of the lactiferous duct tree requires another essential component – mammary epithelial cells extracellular matrix (ECM) which, together with adipocytes, fibroblast, inflammatory cells, and others, constitute mammary stroma. Mammary epithelial ECM mainly contains myoepithelial basement membrane and the connective tissue.

=== Ionization mechanism === In DESI there are two kinds of ionization mechanism, one that applies to low molecular weight molecules and another to high molecular weight molecules. High molecular weight molecules, such as proteins and peptides show electrospray like spectra where multiply charged ions are observed. This suggests desorption of the analyte, where multiple charges in the droplet can easily be transferred to the analyte. The charged droplet hits the sample, spreads over a diameter greater than its original diameter, dissolves the protein and rebounces. The droplets travel to the mass spectrometer inlet and are further desolvated. The solvent typically used for the electrospray is a combination of methanol and water. For the low molecular weight molecules, ionization occurs by charge transfer: an electron or a proton. There are three possibilities for the charge transfer. First, charge transfer between a solvent ion and an analyte on the surface. Second, charge transfer between a gas phase ion and analyte on the surface; in this case the solvent ion is evaporated before reaching the sample surface. This is achieved when the spray to surface distance is large. Third, charge transfer between a gas phase ion and a gas phase analyte molecule. This occurs when a sample has a high vapour pressure.
=== Polymorphisms === A Human Genome Equivalent (HuGE) review in 2004 looked at studies of the connection between genetic mutations affecting leptin regulation and obesity. They reviewed a common polymorphism in the leptin gene (A19G; frequency 0.46), three mutations in the leptin receptor gene (Q223R, K109R, and K656N) and two mutations in the PPARG gene (P12A and C161T). They found no association between any of the polymorphisms and obesity. A 2006 study found a link between the common LEP-2548 G/A genotype and morbid obesity in Taiwanese aborigines, but a 2014 meta-analysis did not, however, this polymorphism has been associated with weight gain in patients taking antipsychotics. The LEP-2548 G/A polymorphism has been linked with an increased risk of prostate cancer, gestational diabetes, and osteoporosis. Other rare polymorphisms have been found, but their association with obesity are not consistent.
=== Early life: 1946–1966 === Bantu Stephen Biko was born on 18 December 1946, at his grandmother's house in Tarkastad, Cape Province (now the Eastern Cape). The third child of Mzingaye Mathew Biko and Alice 'Mamcete' Biko, he had an older sister, Bukelwa, an older brother, Khaya, and a younger sister, Nobandile. His parents had married in Whittlesea, where his father worked as a police officer. Mzingaye was transferred to Queenstown, Port Elizabeth, Fort Cox, and finally King William's Town, where he and Alice settled in Ginsberg township. This was a settlement of around 800 families, with every four families sharing a water supply and toilet. Both Africans and Coloured people lived in the township, where Xhosa, Afrikaans, and English were all spoken. After resigning from the police force, Mzingaye worked as a clerk in the King William's Town Native Affairs Office, while studying for a law degree by correspondence from the University of South Africa. Alice was employed first in domestic work for local white households, then as a cook at Grey Hospital in King William's Town. According to his sister, it was this observation of his mother's difficult working conditions that resulted in Biko's earliest politicisation.
The 2C drugs are metabolized by the monoamine oxidase (MAO) enzymes, including both MAO-A and MAO-B. As a result, they may be potentiated by monoamine oxidase inhibitors (MAOIs), such as phenelzine, tranylcypromine, moclobemide, and selegiline. This may lead to overdose and serious toxicity. There are anecdotal reports of strong potentiation of 2C-B by MAOIs, including hospitalization. There is no known reversal agent for 2C drugs, and medical management for overdose involves treatment of symptoms until toxicity within the body subsides.
== Creation of pan-Slavic languages == Similarities of Slavic languages inspired many to create zonal auxiliary Pan-Slavic languages for all Slavic people to communicate with one another. Several such languages were constructed in the past, but many more were created in the Internet Age. The most prominent modern example is Interslavic.

It is a synthetic seven-amino-acid peptide modeled on alpha-melanocyte-stimulating hormone. It carries two non-natural substitutions and a cyclic bridge that increase its stability relative to the natural hormone. It circulates as a research chemical and is not an approved medicine.
Melanotan-1, also called afamelanotide, is a shorter linear analog with a different amino acid sequence and no lactam ring. It has been evaluated in formal clinical programs, while melanotan-2 has not. The two are distinct molecules and are not interchangeable.
No. Bremelanotide is a related but distinct cyclic peptide that reached approved status for one specific clinical indication. Melanotan-2 is a separate molecule with its own sequence and properties. Shared ancestry in the melanocortin family does not make them the same substance.
No regulatory agency has authorised melanotan-2 as a medicine for any indication. It circulates mainly as a research chemical or through unregulated channels. As a result, identity, purity and content are not independently guaranteed.
