Published pharmacokinetic information is limited and comes mainly from small studies rather than registrational trials. Plasma half-life is usually described as short, on the order of tens of minutes, followed by rapid tissue distribution and clearance of the intact peptide. Metabolites and low concentrations of parent compound have been reported in urine, a detail relevant to anti-doping and forensic testing. Whether repeated exposure changes receptor sensitivity or clearance over time remains an open question. Values differ noticeably between analytical assays, so published numbers should be read as approximate rather than definitive.
Melanotan II is a synthetic cyclic heptapeptide with the sequence Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, corresponding to a molecular formula of C50H69N15O9 and a monoisotopic mass near 1024 daltons. It was designed as a structural analogue of alpha-melanocyte-stimulating hormone, a peptide hormone produced by cleavage of proopiomelanocortin. A lactam bridge between the aspartate and lysine side chains closes the ring, and the C-terminal amide removes a free carboxyl group. Both modifications increase resistance to enzymatic degradation compared with the linear parent hormone. Four substitutions distinguish it from afamelanotide, the linear analogue studied under the name melanotan I.
Receptor-binding studies classify melanotan II as a non-selective melanocortin agonist. It interacts with MC1R, MC3R, MC4R and MC5R, with reported affinities in the low nanomolar range and no strong subtype preference. Activation of MC1R on dermal melanocytes shifts pigment synthesis toward eumelanin, the dark polymer deposited in melanosomes and transferred to keratinocytes. Because the same peptide engages MC4R in the hypothalamus, it also appears in animal work on food intake and erectile response, which is why it is discussed in both pigment and metabolic research. Which receptor populations dominate after systemic exposure in humans is not fully established.
Published human data come mostly from small, short studies rather than large controlled trials. Reported outcomes include increased skin pigmentation and, in some reports, effects on appetite and libido, but sample sizes are small and follow-up is limited. Whether long-term use produces durable pigment changes or adverse effects is not established. Because products sold outside pharmacies are not standardized, the actual content of any given vial is often unknown. Independent testing of such material is uncommon.
Melanotan II is a synthetic cyclic heptapeptide analog derived from the core sequence of alpha-melanocyte-stimulating hormone. Researchers at the University of Arizona synthesized it during the 1980s while studying pigmentation and appetite signaling. The compound is not an approved medicine in any major jurisdiction and appears mainly in laboratory and research-chemical settings. Its structure incorporates a lactam bridge between side chains, which constrains the ring and slows enzymatic breakdown relative to the natural hormone.
Melanotan II binds several melanocortin receptor subtypes rather than a single target. MC1R on melanocytes drives melanin synthesis, while MC3R and MC4R participate in energy balance, appetite, and sexual response pathways. This lack of selectivity explains why reported effects extend beyond skin darkening. Substitutions at positions four and seven, including norleucine and D-phenylalanine, increase potency and resistance to peptidases. Understanding which receptor mediates which effect remains an active area of investigation.

Pharmacologically, melanotan-2 behaves as a non-selective agonist across the melanocortin receptor family. Binding at MC1R on dermal melanocytes promotes eumelanin synthesis, which underlies the tanning response described in early human work. Activity at the centrally expressed MC4R receptor is associated with reported effects on appetite and erectile function. Because the peptide does not discriminate strongly among receptor subtypes, attributing any single observed effect to one receptor pathway is generally not possible without selective antagonists or receptor knockout models.
Research interest has centred on photoprotection and pigmentation, with a smaller body of work on appetite and sexual function. Published human data remain limited to small, frequently uncontrolled studies, and the compound has never received marketing approval from a national medicines regulator. Most laboratory work treats it as a pharmacological tool for probing melanocortin signalling in cell culture or animal models. Whether pigmentation changes observed in people translate into measurable protection against ultraviolet-induced DNA damage remains an open question.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, the naturally occurring peptide involved in pigmentation signalling. Its sequence is conventionally written as Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NH2, with a lactam bridge joining the aspartate side chain to the lysine side chain. The empirical formula is C50H69N15O9 and the monoisotopic mass lies near 1023.5 daltons. N-terminal acetylation and the D-configured phenylalanine both increase resistance to enzymatic breakdown compared with the parent hormone.
Structurally, Melanotan-2 retains the core recognition motif of alpha-melanocyte-stimulating hormone while adding a lactam bridge that links two side chains and constrains the molecule into a ring. This modification lowers susceptibility to enzymatic degradation. The compound acts as an agonist at melanocortin receptors, particularly subtypes associated with melanin production. Because the same receptor family influences several physiological processes, researchers note that its activity is not confined to pigmentation alone. Receptor selectivity continues to be examined in published studies.
Melanotan-2 is a synthetic cyclic heptapeptide designed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation signalling. Its sequence incorporates modified residues that increase potency and extend biological activity relative to the native hormone. The compound binds receptors of the melanocortin family and is examined mainly in laboratory research. It does not occur naturally and exists only as a manufactured chemical entity produced by solid-phase synthesis.
==== Sides are drawn ==== For the latter half of 2003; sides continued to be drawn in the feud as The Source magazine publicly showed support of Ja Rule's in the conflict. The magazine's co-owner at the time Raymond "Benzino" Scott took notice of the ongoing feud as he was involved in a separate public quarrel with Eminem, plausibly displaying his support for Ja Rule in a retaliatory act as Eminem had demonstrated full support for 50 Cent and G-Unit. The toll of the feud began to put itself on display for Ja Rule and Murder Inc by the latter half of 2003; the album Blood in My Eye was released on November 4, 2003 with a heavy emphasis on responding to the feud with both 50 Cent, G-Unit, and Eminem. Despite receiving mixed-to-positive reviews, Blood in My Eye failed to achieve a certification, selling a total of approximately 400,000 copies, indicative of the damage to Murder Inc's public image amidst the feud and their own legal issues with the federal government. Amidst the conflict, Ja Rule still had the support of various high-profile figures within the community, such as Fat Joe and Jadakiss who would go on to appear on Ja Rule's 2004 album R.U.L.E. in the form of the track New York. The album showed a slight return for Ja Rule commercially as it would outsell its predecessor and receive a gold certification, led by the success of New York as a single. The song continued to maintain tensions with 50 Cent and G-Unit as it was perceived to be a subliminal diss track against both 50 Cent and G-Unit member Lloyd Banks, mocking the latter's then-recent single "On Fire".
Carbamazepine is relatively slowly but practically completely absorbed after administration by mouth. Highest concentrations in the blood plasma are reached after 4 to 24 hours depending on the dosage form. Slow release tablets result in about 15% lower absorption and 25% lower peak plasma concentrations than ordinary tablets, as well as in less fluctuation of the concentration, but not in significantly lower minimum concentrations. In the circulation, carbamazepine itself comprises 20 to 30% of total residues. The remainder is in the form of metabolites; 70 to 80% of residues is bound to plasma proteins. Concentrations in breast milk are 25 to 60% of those in the blood plasma. Carbamazepine itself is not pharmacologically active. It is activated, mainly by CYP3A4, to carbamazepine-10,11-epoxide, which is solely responsible for the drug's anticonvulsant effects. The epoxide is then inactivated by microsomal epoxide hydrolase (mEH) to carbamazepine-trans-10,11-diol and further to its glucuronides. Other metabolites include various hydroxyl derivatives and carbamazepine-N-glucuronide. The plasma half-life is about 35 to 40 hours when carbamazepine is given as single dose, but it is a strong inducer of liver enzymes, and the plasma half-life shortens to about 12 to 17 hours when it is given repeatedly. The half-life can be further shortened to 9–10 hours by other enzyme inducers such as phenytoin or phenobarbital. About 70% are excreted via the urine, almost exclusively in form of its metabolites, and 30% via the faeces.
baseline A measure of the gene expression level of a gene or genes prior to a perturbation in an experiment, as in a negative control. Baseline expression may also refer to the expected or historical measure of expression for a gene.
Serine is important in metabolism in that it participates in the biosynthesis of purines and pyrimidines. It is the precursor to several amino acids including glycine and cysteine, as well as tryptophan in bacteria. It is also the precursor to numerous other metabolites, including sphingolipids and folate, which is the principal donor of one-carbon fragments in biosynthesis.
He arranged for the bosses of the leading banks and building societies to tour the area in a coach (they were reluctant until Heseltine's PPS Tim Sainsbury persuaded Robin Leigh-Pemberton of NatWest to come), and asked them to each second a bright young manager to the DOE for a year. Heseltine circulated a 21-page minute to Cabinet on his return, entitled It Took a Riot. He proposed a regional office and a review of the status of the Metropolitan Counties, as well as greater government emphasis on Merseyside in future. He had prepared the ground with a small dinner for Whitehall mandarins including Robert Armstrong (Cabinet Secretary) and Ian Bancroft (Head of the Civil Service). However, Thatcher was not impressed, although she agreed to his appointment as Minister for Merseyside for twelve months. Neither was Keith Joseph (Secretary of State for Industry) nor Howe (Chancellor of the Exchequer), who favoured enterprise zones where businesses would be given favourable tax treatment. Shortly after his appointment as Minister for Merseyside, Heseltine gave his annual party conference speech, in which he condemned talk of repatriation and called for more public spending on inner cities. Although he felt he had taken a risk – the speech was in marked contrast to Norman Tebbit's "On Your Bike" speech a few hours later – he received his usual standing ovation and later recorded that it was the one of his speeches of which he was most proud.

== Epidemiology == Between 0.1% and 0.8% of people are affected. The disease is most common in Northern European countries, and per observations in the UK, is least frequently seen in people of Afro-Caribbean descent. Although the ratio of male to female disease is reportedly 3:1, many rheumatologists believe the number of women with AS is underdiagnosed, as most women tend to experience milder cases of the disease. The majority of people with AS, including 95 per cent of people of European descent with the disease, express the HLA-B27 antigen and high levels of immunoglobulin A (IgA) in the blood. In 2007, a team of researchers discovered two genes that may contribute to the cause of AS: ARTS-1 and IL23R. Together with HLA-B27, these two genes account for roughly 70 percent of the overall number of cases of the disease.
Psilocybe cubensis, commonly known as the magic mushroom, shroom, golden halo, cube, gold cap, or bluestain smoothcap, is a species of psilocybin mushroom of moderate potency whose principal active compounds are psilocybin and psilocin. It belongs to the fungus family Hymenogastraceae and was previously known as Stropharia cubensis. It is the best-known psilocybin mushroom due to its wide distribution and ease of cultivation.
During October and November 2005, Wenger became embroiled in a war of words with Chelsea manager José Mourinho. Mourinho accused Wenger of having an "unprofessional obsession" with Chelsea and labelled him a "voyeur". Mourinho was quoted as saying, "He's worried about us, he's always talking about us – it's [always] Chelsea". Wenger responded by pointing out he was only answering journalists' questions about Chelsea, and described Mourinho's attitude as "disrespectful". Mourinho later apologised and clarified that he regretted his "voyeur" comment; Wenger accepted the apology. In 2014, Mourinho reopened his feud with Wenger by calling him a "specialist in failure". This was in response to Wenger's comments that Chelsea were favourites for the Premier League and managers did not want to take responsibility if they failed to win the title. Wenger said Mourinho's comments were "silly and disrespectful" and had embarrassed Chelsea. When asked if he regretted his remark, Mourinho replied: "You have to ask him if he regrets the comments he made". In October 2014, during a Premier League match between Arsenal and Chelsea, Wenger was involved in a touchline spat with Mourinho. Wenger later apologised for his behaviour. Wenger has directed his anger towards referees when decisions have not gone his team's way. In August 2000, he was charged with "alleged threatening behaviour and physical intimidation" towards fourth official Paul Taylor, after Arsenal's 1–0 defeat at Sunderland on the opening day of the 2000–01 season.
== Tourism == One of Ningxia's main tourist spots is the internationally renowned Xixia Tombs site located 30 km (19 mi) west of Yinchuan. The remnants of nine Western Xia emperors' tombs and two hundred other tombs lie within a 50 km2 (19 sq mi) area. Other famous sites in Ningxia include the Helan Mountains, the mysterious 108 stupas, the twin pagodas of Baisikou and the desert research outpost at Shapotou. A less visited tourist spot in Ningxia is the Mount Sumeru Grottoes (须弥山), which is among the ten most famous grottoes in China.

===== Behavioral treatments ===== A 2018 systematic review and network meta-analysis of 50 trials involving 12 different psychosocial interventions for amphetamine, methamphetamine, or cocaine addiction found that combination therapy with both contingency management and community reinforcement approach had the highest efficacy (i.e., abstinence rate) and acceptability (i.e., lowest dropout rate). Other treatment modalities examined in the analysis included monotherapy with contingency management or community reinforcement approach, cognitive behavioral therapy, 12-step programs, non-contingent reward-based therapies, psychodynamic therapy, and other combination therapies involving these. Additionally, research on the neurobiological effects of physical exercise suggests that daily aerobic exercise, especially endurance exercise (e.g., marathon running), prevents the development of drug addiction and is an effective adjunct therapy (i.e., a supplemental treatment) for amphetamine addiction. Exercise leads to better treatment outcomes when used as an adjunct treatment, particularly for psychostimulant addictions. In particular, aerobic exercise decreases psychostimulant self-administration, reduces the reinstatement (i.e., relapse) of drug-seeking, and induces increased dopamine receptor D2 (DRD2) density in the striatum. This is the opposite of pathological stimulant use, which induces decreased striatal DRD2 density.
== Releases to the environment == PCP has been detected in surface waters and sediments, rainwater, drinking water, aquatic organisms, soil, and food, as well as in human milk, adipose tissue, and urine. As PCP is generally used for its properties as a biocidal agent, considerable concern exists about adverse ecosystem effects in areas of PCP contamination. Releases to the environment are decreasing as a result of declining consumption and changing use methods. However, PCP is still released to surface waters from the atmosphere by wet deposition, from soil by run off and leaching, and from manufacturing and processing facilities. PCP is released directly into the atmosphere via volatilization from treated wood products and during production. Finally, releases to the soil can be by leaching from treated wood products, atmospheric deposition in precipitation (such as rain and snow), spills at industrial facilities, and at hazardous waste sites. After PCP is released into the atmosphere, it decomposes through photolysis. The main biodegradative pathway for PCP is reductive dehalogenation. In this process, the compound PCP is broken down to tetrachlorophenols, trichlorophenols, and dichlorophenols. Another pathway is methylation to pentachloroanisole (a more lipid-soluble compound). These two methods eventually lead to ring cleavage and complete degradation. In shallow waters, PCP is also quickly removed by photolysis. In deep or turbid water processes, sorption and biodegradation take place.
==== Elimination ==== Gabapentin, pregabalin, Baclofen and phenibut are all eliminated renally in the urine. They all have relatively short elimination half-lives, with reported values of 5.0 to 7.0 hours, 6.3 hours, 2.0 to 5.0 hours and 5.3 hours, respectively. Similarly, the terminal half-life of gabapentin enacarbil IR (as active gabapentin) is short at approximately 4.5 to 6.5 hours. Because of its short elimination half-life, gabapentin must be administered 3 to 4 times per day to maintain therapeutic levels. Similarly, pregabalin has been given 2 to 3 times per day in clinical studies. Phenibut, also, is taken 3 times per day. Conversely, gabapentin enacarbil is taken twice a day and gabapentin XR (brand name Gralise) is taken once a day.
== Medical uses == Varicella vaccine is 70% to 90% effective for preventing varicella and more than 95% effective for preventing severe varicella. Follow-up evaluations have taken place in the United States of children immunized that revealed protection for at least 11 years. Studies were conducted in Japan which indicated protection for at least 20 years. People who do not develop enough protection when they get the vaccine may develop a mild case of the disease when in close contact with a person with chickenpox. In these cases, people show very little sign of illness. This has been the case of children who get the vaccine in their early childhood and later have contact with children with chickenpox. Some of these children may develop mild chickenpox also known as breakthrough disease. Another vaccine, known as zoster vaccine, is simply a larger-than-normal dose of the same vaccine used against chickenpox and is used in older adults to reduce the risk of shingles (also called herpes zoster) and postherpetic neuralgia, which are caused by the same virus. The recombinant zoster (shingles) vaccine is recommended for adults aged 50 years and older.
The distinction between Jack and the player has also informed debate over ludonarrative dissonance, a term introduced by game designer Clint Hocking in a 2007 critique of BioShock. Hocking argued that the game permits players to pursue self-interest through its mechanics while obliging Jack to help Atlas through its narrative, creating a conflict between the values expressed through play and those expressed through the story. In a 2026 reassessment, Connor Jackson disputed this reading. Jackson argued that Jack's conditioning provides an in-universe explanation for his obedience to Atlas and cautioned against assuming that the motivations of a controllable avatar must correspond to those of the player. He also emphasized that the rescue-or-harvest decisions concerning the Little Sisters have substantial narrative consequences, and therefore considered BioShock's treatment of agency more complex than a simple contradiction between an inflexible story and flexible gameplay. Jennings identified this disagreement as characteristic of the wider scholarship on BioShock. Although many studies have treated the game as evidence that video-game agency is fundamentally illusory, others have distinguished between different kinds and degrees of agency rather than treating control as either wholly present or absent. She argued that even choices constrained by a designed system can produce meaningful experiences of responsibility, investment, and interpretation, and used the extensive debate surrounding BioShock to argue for a more contingent understanding of agency in games.

No. It is a synthetic analogue carrying four amino acid changes, a lactam ring and an amidated C-terminus. The natural hormone is a linear thirteen-amino-acid peptide processed from proopiomelanocortin.
Afamelanotide is the linear analogue [Nle4-D-Phe7]-alpha-MSH, sometimes called melanotan I, while melanotan II is cyclic and carries three further substitutions. The two are distinct molecules and are not interchangeable in analytical testing.
Binding to the four melanocortin receptor subtypes is well documented in vitro. The relative contribution of each subtype to whole-body effects in humans is far less certain.
No regulator in a major market has approved it for human use. It appears in research settings and in products marketed outside pharmacy channels. Legal status for personal possession varies by country.
